SOEing PCR/Docking Optimization of Protein A-G/scFv-Fc-Bioconjugated Au Nanoparticles for Interaction with Meningitidis Bacterial Antigen

Recent advances in the use of gold nanoparticles (Au NPs)/antibody conjugations nanomedicine have increased need to optimize synthesis conditions and surface functionalization Au NPs. In this study, a home-made Neisseria meningitidis recombinant antibody (scFv-Fc) was developed by connecting fragment crystallizable (Fc) region human with mouse (single-chain variable (scFv)) characterized using SOEing PCR technique. Then, an optimized coating agent for scFv-Fc/Au NP conjugation (i.e., citrate agent) found among three common agents (citrate, allylamine hydrochloride, polyvinyl alcohol) different charges (negative, positive, neutral, respectively). Moreover, stability scFv-Fc/protein A-G presence N-succinimidyl 3-(2-pyridyldithio) propionate (SPDP) linker investigated docking method. It that designed A-G/SPDP/citrate showed bottom-on protein improved NPs, enabling suitable interaction bacterial antigen.
